SEROLOGY SERVICES

smart SEROLOGY …a new approach to serologic monitoring for laboratory animal pathogens using less serum and providing more sensitive and specific results and at a lower cost than traditional ELISA.
The smart SEROLOGY three-tiered approach to serologic testing will
provide the most accurate testing available.

I.  PRIMARY Serologic Testing by MFI (Multiplex Fluorescent Immunoassay) technology

  • MFI replaces the traditional ELISA
  • Provides increased sensitivity and specificity
  • MFI testing allows survival bleeds
    • MFI requires only 0.2 µl of undiluted serum regardless of number of tests requested
      (Recommended submitted sample volume to allow for potential confirmatory testing and ease
      of handling is 100 µl of 1:5 diluted or 20 µl of undiluted serum.)
    • MFI technology makes insufficient serum volumes a thing of the past
  • Cost is lower than traditional ELISA testing
  • MFI results are easy to interpret and correlate with traditional ELISA results

Sample Preparation and Shipping

MFI: We recommend sending 100 µl of serum diluted 1.5 (1 part serum in 4 parts physiologic saline or phosphate-buffered saline). This will allow for testing of our most comprehensive panel as well as sufficient serum for confirmatory testing if required.

ELISA: A minimum of 60 µl of serum diluted 1:5 (1 part serum in 4 parts physiologic saline or phosphate-buffered saline [PBS]) or 12 µl of undiluted serum is required to perform one test. An additional 40 µl of diluted serum (8 µl of undiluted serum) is required for each additional test requested. A further volume of 25% is recommended to allow for retesting of equivocal results.

You may achieve an approximate 1:5 dilution of serum by adding 1 part of whole blood to 2 parts of ambient temperature saline. Refrigerate the diluted blood for 6 to 12 hours, centrifuge at low speed for 5-10 minutes, and recover the 1:5 diluted serum for submission.

Do not submit whole blood; hemolysis will occur during freezing or shipping and may interfere with serologic test performance. Refrain from pooling serum samples, a practice that can yield false negative results by dilution effect.

Serum samples should be shipped frozen using an overnight service. One or two pounds of ice packs are generally adequate depending on the type of container, the number of samples and the ambient temperature. Optimal shipping conditions are achieved when the entire package with its lid open is frozen overnight at or below -20 C. The use of dry ice is not required. If you choose to ship on dry ice, check in advance for special hazardous materials shipping requirements and to be certain that your shipping service will accept dry ice packages.

To expedite testing, preclude leakage, and minimize processing costs, we encourage clients to submit 1:5 diluted serum in RADIL serum vials (provided free of charge on request to clients who pre-dilute their samples) sequentially numbered with an indelible marker. A 5% discount is applied when 10 or more serum samples from one species are submitted for identical testing; a 10% discount for 20 or more.

NOTE: A completed serology case form must be included with each submission specifying the species, serum dilution, and the serology profile or individual test(s) requested. Describe any potential biohazards associated with the samples. Please use a separate serology accession form for each different species of animal and serologic profile or group of test(s) requested.
